چکیده

DPNH-X was prepared as described previously (1) ; the barium salt was converted to the sodium salt before use, and solutions were stored at 20”. Concentrations of DPNH-X were calculated with an extinction coefficient of 13.5 X lo6 sq. cm. mole-’ at 290 rnM (1). Sodium salts of ADP, ATP, ITP, UTP, and crystalline disodium ATP were obtained from the Pabst Laboratories, Milwaukee, Wisconsin. Myokinase was prepared according to the procedure of Colowick and Kalckar (3), and adenylic acid deaminase according to the procedure of Kalckar (4). Alcohol dehydrogenase was obtained from the Worthington Biochemical Corporation, Freehold, New Jersey. Alumina C-r was prepared according to the procedure of Willstatter and Kraut (5).

Chemical properties of 3-substituted pyridine analogues of diphosphopyridine nucleotide.

It has been shown previously that the diphosphopyridine nucleotidase from pig brain is capable of catalyzing an exchange reaction between various substituted pyridine compounds and diphosphopyridine nucleotide to form analogues of diphosphopyridine nucleotide (l-4). The variation in reactivity of the various analogues prepared in this manner indicates the importance of the grouping in position ...

Specific Blood Alcohol Measurements on the Autoanalyzer by Alcohol Dehydrogenase

Introduction m a n y a c c e p t e d chemical methods for blood alcohol involve the oxidation of alcohol after a separation step1’ 2 and the subsequent determination of the excess of oxidant. On the other hand, enzymatic procedures1-4 are based on the conversion of diphosphopyridine nucleotide (DPN) to its reduced form (DPNH), catalysed by alcohol dehydrogenase (ADH) in the presence of ethyl-al...

Automated assay of activities of enzymes involving the diphosphopyridine nucleotide-reduced diphosphopyridine nucleotide reaction.

A continuous flow instrumental system of analysis, first described by Skeggs (l), was previously applied to the determination of phosphohexose isomerase activity and resulted in a method that was approximately eightfold as rapid as the manual procedure (2). The present paper is concerned with the application of this system of analysis to the determination of enzyme activities that utilize the d...

Reevaluation of "ammonium

The intermediate role of hydroxylamine in nitrate assimilation has been suggested by observations that different organisms form ammonia from hydroxylamine in vivo, and that hydroxylamine occurs in nutrient media and cells. In further support are recent investigations describing enzymatic preparations which catalyze the reduction of hydroxylamine to ammonia. Taniguchi et al. (J. Biochem. (Japan)...

Kinetic studies of dogfish liver glutamate dehydrogenase with diphosphopyridine nucleotide and the effect of added salts.

Kinetic studies have been performed with crystalline dogfish liver glutamate dehydrogenase with the diphosphopyridine nucleotides as cofactors. The kinetic constants for the various substrates have been determined. In its sensitivity to guanosine S-triphosphate, adenosine S&phosphate, and excess reduced diphosphopyridine nucleotide the enzyme is similar to that derived from other vertebrates. T...
